Target Information
LSD1 is a histone demethylase that specifically demethylates Lys-4 of histone H3, a marker for epigenetic transcriptional activation, acting as a corepressor. It contains a SWIRM domain, a FAD-binding motif, and an amine oxidase domain. LSD1 functions within histone deacetylase complexes to silence genes by histone demethylation. It oxidizes substrates via FAD to generate imine intermediates, which are subsequently hydrolyzed. LSD1 demethylates both mono- and tri-methylated Lys-4 of histone H3 and may also act on Lys-9, influencing androgen receptor target gene expression. Mutations in LSD1 can lead to developmental defects such as cleft palate and psychomotor retardation.
