Target Information
Replication factor C (RFC) is an essential DNA polymerase accessory protein required for multiple aspects of DNA metabolism, including replication, repair, and telomere maintenance.
RFC functions as a heteropentameric complex that recognizes primer-template DNA, binds to the primer terminus, and loads proliferating cell nuclear antigen (PCNA) onto DNA in an ATP-dependent manner.
All five RFC subunits share conserved RFC box sequences containing nucleotide-binding motifs.
RFC1, RFC2, RFC3, and RFC4 have consensus ATP-binding domains.
RFC2–RFC5 interact with Rad24, whereas RFC1 does not. RFC2, the third-largest subunit, exhibits ATP-binding activity critical for DNA replication and checkpoint control.
