Target Information
Cyclin D1 (PRAD-1, bcl-1)는 세포 주기 조절의 핵심 인자로, CDK4 및 CDK6과 함께 Rb 단백질을 인산화하여 G1/S 전환을 조절합니다. 다양한 인간 종양, 특히 mantle cell lymphoma(MCL)에서 과발현되는 원형암유전자(proto-oncogene)로 알려져 있습니다. Cyclin D1은 세포질 및 핵 내에서 G1 phase 동안 합성되며 성장 인자 자극에 따라 CDK 복합체를 형성합니다. 이 복합체는 Rb 단백질의 성장 억제 기능을 비활성화하고 CDK 억제제(p21Cip1, p27Kip1)를 조절합니다. 성장 인자 자극이 없을 경우 Cyclin D1은 불안정하며, 인산화에 의해 유비퀴틴 매개 분해를 거쳐 G1/S phase 정지를 유도합니다. Cyclin D1 단백질은 세포 주기 동안 단백질 양이 주기적으로 변하는 highly conserved cyclin family에 속하며, CDK4 또는 CDK6의 조절 서브유닛으로 작용합니다. 이 유전자의 돌연변이, 증폭, 과발현은 다양한 종양에서 자주 관찰되며, 종양 발생에 기여할 수 있습니다.
