Product Specific Information

The OKT8 monoclonal antibody recognizes the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as CD8αβ heterodimers or CD8αα homodimers. It is present on most thymocytes and subsets of mature T and NK cells. CD8 binds MHC class I and associates with tyrosine kinase p56lck, playing a role in T-cell development and activation.

Preliminary studies indicate OKT8 does not compete with clones RPA-T8 or HIT8a for binding, suggesting distinct epitopes.
Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for use at 5 µL (0.1 µg)/test. One test = staining a 100 µL cell sample (10⁵–10⁸ cells).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• CellBlox Plus Blocking Buffer (Product # C001T06F01) must be used for optimal staining.
  • Add 5 µL per sample to antibody cocktails before combining with cells.
  • For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

Based on Phiton™ nucleic acid dye structures, engineered for optimized fluorescence with minimal spillover and spread.

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family. It functions as a co-receptor for MHC class I molecules and is essential in T cell signal transduction, differentiation, and activation.

• Expressed mainly on cytotoxic T cells as CD8αβ heterodimers or CD8αα homodimers.
• Also found on NK cells, monocytes, and dendritic cells.
• Binding to MHC-I/peptide complexes triggers Lck kinase recruitment, lymphokine production, motility, and activation of cytotoxic T lymphocytes (CTLs).
• CTL differentiation is enhanced by IL-2, IL-12, and TGF-β1.
