Product Specific Information

The OKT8 monoclonal antibody recognizes human CD8a, a 32–34 kDa cell surface receptor expressed as CD8αβ or CD8αα dimers. CD8a is found on most thymocytes and subpopulations of mature T and NK cells. It binds to MHC class I molecules and interacts with protein tyrosine kinase p56lck, playing a role in T-cell development and activation.

This antibody has been pre-titrated and validated for flow cytometry of normal human peripheral blood cells. Recommended usage is 5 µL (0.1 µg)/test, where one test corresponds to staining a cell sample in 100 µL volume (10⁵–10⁸ cells/test).

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Product # C001T06F01) is required for optimal staining. Add 5 µL per sample before combining with cells.
• For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, providing engineered fluorescence with minimized spectral spillover.

Target Information

CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein receptor of the immunoglobulin family. It functions as a co-receptor with the T cell receptor (TCR) for MHC class I molecules and is essential for T cell activation and cytotoxic T lymphocyte (CTL) function. CD8 is expressed as a heterodimer (CD8αβ) on most T cells and as a homodimer (CD8αα) on subsets of T cells, NK cells, monocytes, and dendritic cells. Engagement with MHC-I/peptide complexes triggers intracellular signaling via Lck kinase, promoting lymphokine production and CTL-mediated immune responses.
