Description:
The OKT8 monoclonal antibody reacts with human CD8a, a 32–34 kDa cell surface receptor expressed on thymocytes, mature T cells, and NK cells. CD8 binds to MHC class I and associates with protein tyrosine kinase p56lck, playing a role in T-cell development and activation.

Preliminary testing shows that OKT8, RPA-T8, and HIT8a clones do not compete for binding to human peripheral blood leukocytes, suggesting distinct epitopes.

Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometry using normal human peripheral blood cells at 5 µL (0.1 µg)/test.
A test is defined as the amount required for staining a 100 µL cell sample (10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
• Use CellBlox Plus Blocking Buffer (C001T06F01) for optimal staining.
  • Add 5 µL per sample to antibody cocktails before combining with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample.

Based on Phiton™ technology utilizing nucleic acid dye structures for engineered fluorescent signatures with reduced spectral spillover.

Target Information

CD8 (Cluster of Differentiation 8):
A type I transmembrane glycoprotein of the immunoglobulin receptor family involved in T-cell signaling, differentiation, and activation.

• Expressed mainly on T cells as a CD8αβ heterodimer or CD8αα homodimer.
• Functions as a co-receptor with TCR for MHC class I molecules.
• Also found on subsets of NK cells, monocytes, and dendritic cells.
• Engagement with MHC-I/peptide complexes triggers Lck recruitment, lymphokine production, and cytotoxic T lymphocyte (CTL) activation.
• CTL differentiation is enhanced by IL-2, IL-12, and TGF-β1.
