Product Specific Information

The 205yekta antibody reacts with mouse CD205, also known as DEC-205 (dendritic and epithelial cells, 205 kDa).
CD205 is an integral membrane glycoprotein involved in antigen uptake, trafficking, and presentation that improves induction of antigen-specific T cell immunity. It is highly expressed by CD8+ dendritic cells and also expressed at varying levels by bone marrow Gr1+ cells, Langerhans cells, BMDCs, and thymic epithelial cells.

Important Notes:

• CD205 binds PE-Cy5.5 antibody and streptavidin conjugates; avoid using these formats on CD205+ dendritic cells.
• Each product includes 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.
• Applications Reported: Flow cytometric analysis.
• Applications Tested: ≤0.2 µg/test (100 µL final volume, 10⁵–10⁸ cells/test).
• Antibody should be titrated for optimal performance.

Compatibility:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

Blocking Buffer Usage:

• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL per sample to antibody cocktails before combining with cells.
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Technology:

• NovaFluor conjugates use Phiton™ technology with engineered nucleic acid dye structures for optimized spectral performance.
• Excitation: 552 nm; Emission: 663 nm; Laser: 561 nm (Yellow).
• Internal testing shows strain-dependent non-specific staining: B cells in SJL mice show non-specific staining, not observed in BALB/c or C57BL/6.

CD205 (LY25, Lymphocyte Antigen 75) is an integral membrane glycoprotein involved in antigen uptake, trafficking, and presentation that enhances antigen-specific T cell immunity.
Highly expressed by CD8+ dendritic cells, bone marrow Gr1+ cells, Langerhans cells, BMDCs, and thymic epithelial cells.
Acts as an endocytic receptor directing captured antigens to specialized processing regions and modulates B lymphocyte proliferation.
Associated diseases include pneumonic plague and lipid pneumonia.
