Target Information
Paraoxonase-1 (PON1) is a calcium-dependent ester hydrolase associated with apoA-I in HDL. PON1 prevents LDL oxidation and formation of atherogenic oxidized-LDL. Polymorphisms of the human PON1 gene correlate with coronary artery disease, suggesting a genetic link. PON1 activity is lower in individuals prone to atherosclerosis (e.g., diabetes, familial hypercholesterolemia, renal disease).
PON2 is an intracellular protein that protects cells against oxidative damage. PON3 is similar to PON1 in function but differs in substrate specificity. Both PON1 and PON3 are implicated in reducing the risk of coronary artery disease and atherosclerosis. Human PON3 shares three conserved cysteine residues with PON1, important for in vivo activity.
PON1-based catalytic scavengers may offer a safe and effective approach to organophosphate detoxification.
