Product Specific Information
The RM4-5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by thymocytes, mature T cells, and dendritic cells. CD4 binds to MHC class II on antigen-presenting cells and plays a key role in T cell development and function. RM4-5 binding is blocked by GK1.5.
Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤1.0 µg/test (100 µL final volume). Optimal titration recommended.
Brilliant Violet™ 786 (BV786):
- Tandem dye emitting at 786 nm
- Requires violet (405 nm) laser
- Sensitive to light; protect from photo-oxidation
Staining Buffer Recommendation:
Use Super Bright Complete Staining Buffer (SB-4401) or Brilliant Stain Buffer™ (00-4409-75) when combining multiple polymer dyes to minimize non-specific interactions.
Fixation Compatibility:
Samples can be stored in IC Fixation Buffer (00-8222) or Fix/Lyse Solution (00-5333) for up to 3 days at 4°C in the dark. BV786 is not compatible with methanol-based fixation.

Target Information
The CD4 antigen recognizes MHC class II molecules and serves as a co-receptor for HIV. Expressed mainly in T helper cells, as well as monocytes, macrophages, and dendritic cells. CD4 expression is found in thymus, lymph nodes, tonsils, spleen, and various non-lymphoid tissues. It assists in T-cell activation via association with Lck kinase and may play a role in neuronal damage in immune-related CNS diseases.
