Product Specific Information

This fc23cpg monoclonal antibody reacts with the p19 subunit of mouse IL-23, a heterodimeric cytokine composed of p40 and p19 subunits. IL-23 signals through a heterodimeric receptor expressed on activated T cells, macrophages, and dendritic cells, sharing IL-12Rβ1 with IL-12 but possessing a unique IL-23R chain. Binding of IL-23 activates Jak/STAT pathways similar to IL-12, but induces distinct immune responses such as RORγt expression and Th17 differentiation.

Dendritic cells and macrophages produce IL-23 in response to TLR2, TLR4, TLR8 ligands, and Dectin-1 agonists. Expression levels vary depending on donor and stimulation conditions. Recent studies indicate that the p19 subunit may exist independently of p40 association.

Target Information

IL-23 is a heterodimeric cytokine consisting of the IL-12 p40 subunit linked to p19. The p19 subunit alone is biologically inactive. IL-23 interacts with IL-12Rβ1 and a unique IL-23R subunit containing a STAT4-binding domain. It is secreted by activated dendritic cells and exhibits distinct biological activities compared to IL-12.

In mice, IL-23 induces proliferation of memory T cells and production of IL-17, but not IFN-γ. In humans, IL-23 promotes memory T cell proliferation and moderate IFN-γ production, showing partially overlapping functions with IL-12.
