Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase (PTP) expressed in all nucleated hematopoietic cells, comprising approximately 10% of all surface proteins in lymphocytes. CD45 is absent on non-hematopoietic cell lines and tissues. It plays a crucial role in lymphocyte development and antigen signaling, regulating Src-family kinases. CD45 exists as multiple isoforms due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic domains. CD45RA is one isoform with restricted expression among lymphoid subtypes. CD45 isoforms differ in their ability to translocate into glycosphingolipid-enriched membrane domains, depending on cell type and physiological state. It is an essential regulator of T- and B-cell receptor signaling, suppressing JAK kinases and modulating cytokine receptor signaling. CD45 also promotes cell survival through integrin-mediated signaling, DNA fragmentation during apoptosis, and regulation of various immunological functions.
