Target Information
The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. It mediates the physiological effects of progesterone, which plays a central role in reproductive events associated with the establishment and maintenance of pregnancy.
PR exists as two main isoforms:
- Form A (94 kDa)
- Form B (120 kDa)
These isoforms arise from alternative translation initiation sites. PR-B is the transcriptionally active form responsible for activating genes related to endometrial maintenance, pregnancy support, and ovulation inhibition. PR-A differs from PR-B by a 165 amino acid deletion at the N-terminus, exposing a 140 amino acid inhibitory domain that represses steroid hormone transcriptional activity.
In its inactive state, the progesterone receptor forms a multiprotein complex including heat shock proteins and immunophilins. Upon hormone binding, the receptor undergoes conformational change, dimerizes, and binds to progesterone response elements (PRE) to activate transcription. A null mutation in the PGR gene results in pleiotropic reproductive abnormalities.
