Product Specific Information

Description:
The PK136 monoclonal antibody reacts with mouse NK1.1, an antigen expressed by natural killer (NK) cells and a subset of T cells in NK1.1-positive mouse strains (e.g., C57BL, NZB). Common strains such as BALB/c, SJL, AKR, CBA, C3H, and A do not express NK1.1. For these strains, use the DX5 monoclonal antibody (Product #14-5971).

Simultaneous staining of C57BL/6 spleen cells with PK136 and DX5 reveals coexpression of both markers by most cells, with minor DX5⁺PK136⁻ and DX5⁻PK136⁺ subsets.

Applications Reported:
Flow cytometric analysis.

Applications Tested:
Tested by flow cytometry using C57BL/6 mouse splenocytes. Recommended ≤0.25 µg/test (100 µL final volume). Cell count range: 10⁵–10⁸ cells/test. Optimize antibody titration for best performance.

Target Information

NK1.1 (KLRB1, NKR-P1A, CD161):
A receptor expressed on NK cells and some T cells, belonging to the C-type lectin superfamily. It contains extracellular lectin-like motifs, a transmembrane domain, and a cytoplasmic domain, classifying it as a type II membrane protein. The receptor recognizes LLT1 (Lectin Like Transcript-1) as its functional ligand and plays a role in NK cell activation and regulation.
