Target Information
BACE1 (beta-secretase)는 알츠하이머병(AD)의 아밀로이드 베타 펩타이드(Abeta) 생성에 관여하는 주요 효소입니다. 일부 조기 발병 가족성 AD는 APP(amyloid precursor protein)의 ‘Swedish’ 돌연변이로 인해 BACE1 절단이 증가하는 것으로 알려져 있습니다. 이러한 유전적·병리학적 근거를 바탕으로 BACE1 및 γ-secretase 억제제를 이용한 치료 연구가 진행되고 있습니다.
APP는 β-secretase에 의해 먼저 절단되어 용해성 단백질 유도체와 막 결합형 99-아미노산 C-말단 조각(C99)을 형성하며, 이 C99는 γ-secretase의 기질로 작용하여 4 kDa 크기의 amyloid-β 펩타이드를 생성합니다. β-secretase는 여러 연구 그룹에 의해 독립적으로 확인되어 BACE(Asp2)로 명명되었으며, 새로운 막관통형 아스파르틸 프로테아제로 APP와 공위치합니다.
