Target Information
IkB-alpha는 NF-κB 활성을 억제하는 40 kDa 단백질로, 세포질 내에서 NF-κB 이합체와 결합하여 핵 내 이동을 방지합니다. TNF-α, IL-1β, PMA, 성장인자, LPS, UV 조사, 바이러스 감염 등 다양한 자극에 의해 IkBα가 분해되면 NF-κB가 활성화됩니다.
인간에서는 IkBα 유전자가 14번 염색체 q arm에 위치하며, 활성화 과정에서 IkB는 특정 세린 잔기(Ser32, Ser36)에서 인산화되어 분해됩니다. 이 반응은 IkB kinase alpha (IKKα)에 의해 매개되며, NF-κB 유도 키나제(NIK)가 이를 조절합니다. IKK 복합체는 IKKα, IKKβ, IKKγ의 세 가지 서브유닛으로 구성되며, 각각 IkB 인산화에 필수적인 역할을 합니다.
