Target Information
PAI1 (plasminogen activator inhibitor 1) belongs to the serine protease inhibitor superfamily and is the principal inhibitor of tissue-type and urokinase-type plasminogen activators (tPA and uPA). Platelets are the main source of circulating PAI1, but it is also synthesized and secreted by fibroblasts, smooth muscle cells, endothelial cells, hepatocytes, and inflammatory cells.
Expression of PAI1 can be regulated by growth factors, cytokines, hormones, inflammatory factors, glucose or lipid metabolites, and environmental factors. Elevated PAI1 levels are associated with thrombosis, obesity, thrombophilia, and metabolic syndrome.
Defects in PAI1 can cause abnormal bleeding or plasminogen activator inhibitor-1 deficiency (PAI-1 deficiency). PAI1 mediates inhibition of fibrinolysis and may promote neuronal survival. Alternatively spliced transcript variants encoding different isoforms have been identified.
