Target Information

GRP78 is a 78 kDa glucose-regulated protein belonging to the heat shock protein family (HSP70). It is a resident protein of the endoplasmic reticulum (ER) and transiently associates with newly synthesized secretory and membrane proteins.
GRP78 interacts with mutant and misfolded proteins, marking them for degradation. The conserved C-terminal sequence Lys-Asp-Glu-Leu (KDEL) is recognized by the KDEL receptor, ensuring recycling of escaped ER proteins back to the ER.
It plays a critical role in protein folding, degradation of misfolded proteins, and maintaining cell homeostasis to prevent apoptosis.
GRP78 levels serve as a biomarker of hypoglycemia and provide neuroprotection against glutamate and oxidative stress. Reduced GRP78 expression has been observed in Alzheimer’s disease and is associated with PS1 gene mutations.
Induction of GRP78 has also been linked to the development of resistance to antitumor drugs.
