Target Information
Dapper homolog 1 (DACT1), an ortholog of Xenopus Dpr, antagonizes Wnt signaling by promoting Dvl degradation through a lysosome inhibitor-sensitive and proteasome inhibitor-insensitive mechanism. It acts as an interacting protein for Dvl, with interaction occurring between the DEP domain of Dvl and the central/C-terminal regions of DACT1. The human DACT1 gene is located on chromosome 14q23.1. This protein regulates β-catenin degradation, enhancing transcription of Wnt pathway target genes, and is essential for normal notochord formation. It is downregulated in hepatocellular carcinoma and expressed in liver and other tissues.
