Target Information

Na⁺/H⁺ exchangers (NHE) are plasma membrane intrinsic proteins that mediate the exchange of Na⁺ and H⁺ ions in various tissues. They play a crucial role in regulating intracellular pH, trans-cellular Na⁺ absorption, cell volume regulation, and possibly cell proliferation.

These exchangers are implicated in several pathophysiological conditions including hypertension, organ growth and hypertrophy, cancer regression, and renal or intestinal disorders.

At least seven NHE isoforms (NHE1–7) have been cloned, sharing similar primary structures with 10–12 predicted transmembrane domains. The C-terminal domain is predicted to be intracellular.

NHE7 (human, 725 aa, chromosome Xp11.4) is ubiquitously expressed, primarily localized to the trans-Golgi network, and mediates Na⁺ or K⁺ influx in exchange for H⁺. It shows approximately 70% similarity to NHE6 and about 25% homology with other NHEs.
