Product Specific Information

The H57-597 monoclonal antibody reacts with the beta chain of mouse TCR. TCR beta is expressed by thymocytes in a developmentally regulated manner and by most peripheral T cells. Crosslinking of the TCR complex with H57-597 induces activation, proliferation, or apoptosis depending on assay conditions. It serves as a phenotypic marker for TCR beta-expressing T cells.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Tested: Mouse thymocytes and splenocytes (≤ 0.8 µg/test)

A test is defined as the amount of antibody staining a cell sample in 100 µL final volume. Recommended to titrate carefully for optimal performance.

Compatibility Notes:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):
Required for optimal staining with NovaFluor conjugates. Add 5 µL per sample before combining with cells. For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology based on nucleic acid dye structures, engineered for optimized fluorescence with minimized spillover and spread.

Target Information

T cell receptors (TCRs) discriminate foreign from self-peptides presented by MHC class II molecules, essential for adaptive immunity. Self-peptide recognition is linked to autoimmune diseases, while mutant peptides relate to tumors. Superantigen engagement can lead to toxic shock syndrome. Autoantibodies to TCR V beta segments have been found in rheumatoid arthritis and lupus, potentially modulating inflammatory responses.

Most human T cells express TCR alpha-beta with CD4 or CD8. A small subset lacks both (double negative). Gamma-delta T cells, found mainly in epithelia, exhibit limited diversity and unique antigen recognition, with subsets showing antitumor and immunoregulatory roles.
