Target Information

Retinoic acid-inducible gene I (RIG-I) is a pattern recognition receptor (PRR) involved in the detection of viral double-stranded RNA (dsRNA). Together with MDA5, RIG-I activates innate immune responses by recognizing viral RNA. Both proteins are RNA helicases with overlapping and distinct functions.
RIG-I is activated by short dsRNAs lacking a 5′-triphosphate end, while MDA5 responds to long dsRNAs. Upon activation, RIG-I signals through IPS-1, leading to activation of transcription factors NF-κB and IRF-3 and inducing apoptosis, cytokine signaling, and inflammation.
RIG-I plays a critical role in immune responses against influenza A/B, respiratory syncytial virus, paramyxoviruses, Japanese encephalitis virus, and West Nile virus. MicroRNA-146a negatively regulates RIG-I-dependent antiviral signaling by targeting TRAF6, IRAK1, and IRAK2. Recent studies also suggest RIG-I involvement in cytosolic DNA detection via RNA polymerase III activity.
