Description:
The monoclonal antibody miap301 reacts to CD47, also known as integrin-associated protein (IAP) and neurophilin. CD47 is a glycosylated five-transmembrane protein with a small cytoplasmic domain. It is involved in adhesion through interactions with SIRP (signal regulator protein) and is associated with β3 integrins (CD51/CD61, CD41/CD61). CD47 also serves as a receptor for thrombospondin.
Expression is found in most hematopoietic cells (T, B cells, monocytes, platelets, erythrocytes) and non-hematopoietic cells. No crossreactivity is seen against human CD47.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes at ≤1 µg/test (100 µL final volume). Optimize titration for best results.
Light Sensitivity: This tandem dye is sensitive to photo-induced oxidation. Protect from light.
Fixation: Samples can be stored in:
- IC Fixation Buffer (Product #00-8222)
- 1-step Fix/Lyse Solution (Product #00-5333)
for up to 3 days at 4°C in the dark with minimal impact on brightness or FRET efficiency.

Target Information
CD47 (integrin-associated protein, IAP) is a ubiquitously expressed cell surface transmembrane glycoprotein that interacts with several integrins and regulates their functions. It modulates signaling pathways such as activation of heterotrimeric G proteins and increases intracellular calcium upon adhesion.
CD47 serves as a receptor for the C-terminal domain of thrombospondin and plays roles in membrane transport and signal transduction. It has broad tissue distribution and acts as a ligand for CD172a (SIRPα), preventing phagocytosis of CD47-positive cells. The CD47-CD172a system also influences cell migration, B cell adhesion, T cell activation, chondrocyte mechanotransduction, and neuronal development, especially in synapse-rich brain and retinal regions.
