Target Information

Adenosine Deaminase RNA Specific (ADAR) catalyzes the hydrolytic deamination of adenosine to inosine in double-stranded RNA (dsRNA), referred to as A-to-I RNA editing.
This process may affect gene expression and function in various ways, including:

  • Altering mRNA translation by changing codons and amino acid sequences
  • Modifying pre-mRNA splicing through splice site recognition changes
  • Influencing RNA stability and nuclease recognition
  • Affecting viral RNA replication and RNA structure-dependent processes such as microRNA production or protein-RNA interactions

ADAR edits both viral and cellular RNAs, either at multiple sites (hyper-editing) or specific sites (site-specific editing).
Cellular RNA substrates include BLCAP, GRIA2, HTR2C, and GABRA3, where site-specific editing can alter protein function.

Viral RNA substrates include HCV, VSV, MV, HDV, and HIV-1.
ADAR exhibits proviral or antiviral effects depending on the virus and mechanism (editing-dependent or -independent).
For example:

  • Inhibits HCV replication via RNA editing
  • Enhances MV, VSV, and HIV-1 replication via suppression of EIF2AK2/PKR activation
  • Promotes HIV-1 infectivity by editing adenosines in viral RNA regions
  • Enhances HDV replication by editing an amber stop codon to a tryptophan codon, enabling synthesis of L-HDAg

However, excessive ADAR1 expression can inhibit HDV replication.
