Target Information

Adenosine Deaminase RNA Specific (ADAR) catalyzes the hydrolytic deamination of adenosine to inosine in double-stranded RNA (dsRNA), a process known as A-to-I RNA editing.
This modification can influence gene expression and function by:

  • Changing codons and altering protein amino acid sequences
  • Modifying splice site recognition sequences in pre-mRNA
  • Affecting RNA stability and nuclease recognition
  • Influencing RNA virus genome stability and replication
  • Regulating microRNA production and RNA–protein interactions

ADAR edits both viral and cellular RNAs, either at multiple sites (hyper-editing) or specific sites (site-specific editing).
Known cellular RNA substrates include BLCAP, GRIA2, HTR2C, and GABRA3, where editing leads to amino acid substitutions and altered protein function.

Viral RNA substrates include HCV, VSV, MV, HDV, and HIV-1.
ADAR exhibits proviral or antiviral effects depending on the virus and mechanism, which may be editing-dependent or independent. For example:

  • Inhibits HCV replication via RNA editing
  • Enhances replication of MV, VSV, and HIV-1 through suppression of EIF2AK2/PKR activation
  • Stimulates HIV-1 infectivity by editing adenosines in the 5′UTR and coding regions of Rev and Tat
  • Enhances HDV replication by editing the amber/W site to produce the large delta antigen (L-HDAg)

However, high levels of ADAR1 can inhibit HDV replication.
