Target Information

Glutathione peroxidases (Gpxs) are ubiquitously expressed enzymes that catalyze the reduction of hydrogen and organic hydroperoxides using glutathione. Multiple isoforms exist with distinct structures and localizations:

• GPx1 (cGPx): Cytosolic/mitochondrial, selenium-dependent enzyme, first discovered in the GPx family.
• GPx2 (GI-GPx): Expressed in gastrointestinal epithelium.
• GPx3 (pGPx): Extracellular plasma enzyme, mainly expressed by the kidney and secreted into circulation.
• GPx4 (PH-GPx): Found in most tissues, reduces hydroperoxides in membranes and lipoproteins.

All mammalian GPx family members, except Cys-containing GPx3 and epididymis-specific GPx5, possess selenocysteine at the active site.
