Target Information
GRP78 is a 78 kDa glucose regulated protein belonging to the heat shock protein family (including HSP70). It resides in the endoplasmic reticulum (ER) and transiently associates with newly synthesized secretory and membrane proteins.
GRP78 also interacts with mutant and misfolded proteins, marking them for degradation. The conserved sequence Lys-Asp-Glu-Leu (KDEL) at the C-terminus is recognized by the KDEL receptor, ensuring recycling of ER proteins.
It plays a key role in protein folding and degradation of misfolded proteins through interaction with DNAJC10, maintaining cell homeostasis and preventing apoptosis.
GRP78 levels serve as biomarkers of hypoglycemia and have neuroprotective roles under oxidative stress. Decreased GRP78 expression is observed in Alzheimer’s disease and associated with presenilin-1 mutations. Induction of GRP78 is linked to antitumor drug resistance.
