The Human 6Ckine (CCL21) Simplex ProcartaPlex Kit measures 6Ckine (CCL21) protein and is designed to be combinable with other Simplex kits, allowing creation of custom multiplex panels utilizing Luminex xMAP technology for protein detection and quantitation. When combining multiple Simplex kits (not using a pre-configured Multiplex Panel), only one buffer kit (sold separately) is required per assay plate regardless of plex size.

About ProcartaPlex Assays for the Luminex Platform

ProcartaPlex immunoassays are based on sandwich ELISA principles, using two highly specific antibodies binding to different epitopes of one protein to quantitate multiple protein targets simultaneously on a Luminex instrument. These assays require as little as 25 μL of plasma or serum, or 50 μL of cell culture supernatant, and yield results within four hours.

ProcartaPlex assays utilize Luminex xMAP (multianalyte profiling) technology for simultaneous detection and quantitation of up to 80 protein targets in a single sample. The Luminex beads are internally dyed with precise proportions of red and infrared fluorophores to create spectrally unique signatures identifiable by Luminex detection systems (e.g., Luminex 200, FLEXMAP 3D, MAGPIX).

Similar to a sandwich ELISA, matched antibody pairs identify the target protein. In multiplexed assays, each spectrally unique bead is labeled with antibodies specific for a single protein target, and bound proteins are detected using biotinylated antibodies and streptavidin–R-phycoerythrin (RPE). The bead-based capture system allows simultaneous analysis of multiple targets in a single well.

Unlike traditional ELISA, the capture antibody in ProcartaPlex assays is conjugated to a bead rather than adsorbed to a microplate well, allowing reagents to remain in free solution. Detection instruments such as the Luminex 200 use dual lasers to distinguish bead signatures and quantify RPE fluorescence proportional to protein amount. ProcartaPlex multiplex assays enable profiling of more targets using less sample in the same time as a standard ELISA.

ProcartaPlex multiplex panels are available in multiple formats across six species (human, mouse, rat, nonhuman primate, porcine, and canine).
