Target Information

Immunophilins are soluble cytosolic receptors that bind immunosuppressant agents cyclosporin A (CsA) or FK506. Proteins binding FK506 are FK506 Binding Proteins (FKBPs), while those binding CsA are cyclophilins (CyP). Both CyP:CsA and FKBP:FK506 complexes inhibit calcineurin, a calcium and calmodulin-dependent protein phosphatase involved in T-cell activation, explaining the immunosuppressive mechanism of CsA and FK506.
Immunophilins act as peptidyl prolyl cis-trans-isomerases (PPIase), accelerating protein folding in vivo and in vitro. Within the cyclophilin family, CyPA (also known as CyP-18) is the most abundant and ubiquitous member, found in all vertebrate tissues including T-cells.
