Product Specific Information

This antibody was cross-adsorbed to phospho-tyrosine coupled to agarose then affinity purified using phospho-IkappaBalpha (Tyr-305) peptide (without carrier).
The antibody detects a 38 kDa protein on SDS-PAGE immunoblots of A431 and Jurkat cells treated with pervanadate, but does not detect this band in control cells.


Target Information

IkB-alpha is a 40 kDa protein that functions to inhibit NF-κB activity through protein-protein interaction between IkB proteins and NF-κB dimers in the cytosol.
This interaction masks the nuclear localization sequence of NF-κB, preventing its translocation to the nucleus.
Various stimuli such as TNF-α, IL-1β, chemokines, PMA, growth factors, LPS, UV irradiation, viral infection, and other stresses can induce degradation of IkB-alpha, thereby activating NF-κB.
In humans, the gene is located on the q arm of chromosome 14.
Activation of NF-κB requires phosphorylation of IkB on specific serine residues (Ser32 and Ser36), leading to its degradation.
IkB kinase alpha (IKKα, also known as CHUK) interacts with IkB-alpha and specifically phosphorylates it at these sites.
IKKα is critical for NF-κB activation in response to proinflammatory cytokines, and its activity is regulated by NF-κB inducing kinase (NIK).
The functional IKK complex contains three subunits: IKKα, IKKβ, and IKKγ, each contributing to IkB phosphorylation.
