Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family. It plays a critical role in signal transduction, T-cell differentiation, and activation. CD8 is primarily expressed on T cells as a disulfide-linked heterodimer (CD8α/CD8β) acting as a co-receptor with TCR for MHC-I molecules, while CD4 serves as a co-receptor for MHC-II. CD8α is essential for MHC-I binding and is also found as a homodimer on subsets of T cells, NK cells, monocytes, and dendritic cells. Engagement of MHC-I/peptide complexes recruits Lck kinase, leading to lymphokine production, motility, and CTL activation. Activated CTLs are vital for pathogen and tumor cell clearance. Differentiation of naive CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
