Target Information

The SMAD1 gene encodes a member of the SMAD family, which are signal transducers and transcriptional modulators similar to the Drosophila gene mothers against decapentaplegic (Mad) and the C. elegans gene Sma. SMAD proteins mediate multiple signaling pathways, particularly those of bone morphogenetic proteins (BMPs), which regulate cell growth, apoptosis, morphogenesis, development, and immune responses. Upon BMP ligand binding, SMAD1 is phosphorylated and forms a complex with SMAD4, enabling transcriptional regulation. It is also targeted by SMAD-specific E3 ubiquitin ligases such as SMURF1 and SMURF2, leading to ubiquitination and proteasomal degradation. Alternatively spliced transcript variants encoding the same protein have been observed.
