SIGIRR (single Ig IL-1R-related molecule)는 interleukin 1 receptor (IL-1R) 초가족의 새로운 구성원입니다. IL-1R 및 Toll 초가족의 보존된 모티프를 공유하지만, 구조적·기능적으로 구별됩니다.
SIGIRR은 세포외 영역에 단일 Ig 도메인을 가지며, IL-1R 계열의 세 Ig 폴드와는 다릅니다. 긴 세포질 도메인은 Drosophila Toll의 구조를 연상시키지만, 펩타이드 서열은 IL-1RI와 더 유사합니다.
SIGIRR의 세포내 도메인을 IL-1R의 세포외 도메인과 융합한 키메라 단백질은 NFκB를 활성화하지 않았으며, 이는 다른 IL-1R 관련 단백질과는 대조적입니다. 계통학적 분석에 따르면 SIGIRR은 포유류에서 MyD88의 기능적 근연체로 추정됩니다.
