Target Information

The Calpain family of proteins are calcium-regulated thiol proteases with broad endopeptidase activity. Calpain small subunit 2 (CSS2 or CAPNS2) is a calcium-dependent protease expressed ubiquitously in the cytoplasm. It forms a heterodimer with a large subunit, catalyzing proteolysis of proteins involved in cytoskeletal remodeling and signal transduction. CSS2 acts as a chaperone for the large subunit, maintaining its folding and conformation. Upon calcium binding, CSS2 dissociates from the complex, activating the large subunit and translocating to the cell membrane. Defects in the CSS2 gene cause abnormal calpain activity and impaired fetal development, indicating its essential role in growth.
