Target Information

F-Box and WD Repeat Domain Containing 7 (FBXW7) is a member of the F-box protein family characterized by an approximately 40 amino acid motif, the F-box. These proteins form part of the SCF (SKP1–Cullin–F-box) ubiquitin ligase complex, functioning in phosphorylation-dependent ubiquitination.
F-box proteins are classified into three types:

  • Fbws: Contain WD-40 domains
  • Fbls: Contain leucine-rich repeats
  • Fbxs: Contain other interaction modules or no recognizable motifs

FBXW7, also known as FBX30, belongs to the Fbws class and includes 7 tandem WD40 repeats. It binds directly to cyclin E and likely targets it for ubiquitin-mediated degradation. Mutations in this gene have been detected in ovarian and breast cancer cell lines, suggesting its involvement in cancer pathogenesis. Multiple transcript variants encoding different isoforms have been identified.
