CD30 (Ki-1, TNF Receptor Superfamily Member 8)
CD30 is a type I transmembrane glycoprotein of the TNF receptor superfamily. It was originally identified as a cell surface antigen of Hodgkin’s and Reed-Sternberg cells using monoclonal antibody Ki-1. The ligand for CD30 is CD30L (CD153), and their interaction mediates various cellular processes such as proliferation, activation, differentiation, and apoptosis.

CD30 plays a critical role in the pathophysiology of Hodgkin’s disease and other CD30⁺ lymphomas. It functions as a costimulatory molecule in thymic negative selection. CD30 is expressed not only on Hodgkin’s and Reed-Sternberg cells but also in certain non-Hodgkin’s lymphomas, virus-infected T and B cells, and activated normal T and B cells.

In T cells, CD30 expression is found on subsets producing Th2-type cytokines and on CD4⁺/CD8⁺ thymocytes co-expressing CD45RO and IL4 receptor. The soluble form (sCD30) serves as a marker of Th2 immune response. CD30 interacts with TRAF2 and TRAF5 to activate NF-κB signaling and acts as a positive regulator of apoptosis, limiting autoreactive CD8⁺ effector T cell proliferation and contributing to immune regulation.

Two alternatively spliced transcript variants encoding distinct isoforms have been reported. CD30 is expressed by mononuclear cells in Hodgkin’s lymphoma, Reed-Sternberg cells, and most Anaplastic Large Cell Lymphomas (ALCL), as well as embryonal carcinomas.
