Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or pyrimidine nucleotide starvation. Upon stress, p53 activates transcription of specific genes such as p21waf1, leading to G1 or G2/M arrest. Structurally, p53 consists of an N-terminal transactivation domain, a central DNA-binding domain, an oligomerisation domain, and a C-terminal regulatory domain.

Phosphorylation sites on p53, including Ser15 and Ser392, are critical for activation, stabilization, and tumor association. p53 plays roles in preventing proliferation of damaged cells, aging, oxidative stress response, and cognitive function regulation. Mutations in p53, common in human cancers, disrupt DNA binding and tumor suppressor activity. TP53 gene alterations occur as both somatic and germline mutations, including in Li-Fraumeni syndrome.
