Target Information
Component of the 20S core proteasome complex involved in the proteolytic degradation of most intracellular proteins. This complex plays essential roles by associating with different regulatory particles.
When associated with two 19S regulatory particles, it forms the 26S proteasome, participating in ATP-dependent degradation of ubiquitinated proteins. The 26S proteasome maintains protein homeostasis by removing misfolded or damaged proteins and proteins no longer needed.
Association with PA200 or PA28 enables ubiquitin-independent protein degradation, required in pathways such as spermatogenesis (20S–PA200 complex) or generation of MHC class I antigenic peptides (20S–PA28 complex).
Inhibits HIF-1A transactivation under normoxic and hypoxic conditions. Interaction with EMAP2 enhances proteasome-mediated HIF-1A degradation under hypoxia. Plays roles in hepatitis C virus translation and androgen receptor nuclear translocation, enhancing androgen-mediated transactivation.
