Product Specific Information

The MEL-14 monoclonal antibody reacts with mouse CD62L, a 76 kDa member of the selectin family. CD62L is expressed by neutrophils, monocytes, and subsets of T, B, and NK cells and binds glycosylated, fucosylated, sulfated sialylated glycoproteins such as CD34, glycam-1, and MAdCam-1. These interactions mediate lymphocyte rolling on activated endothelium and homing to high endothelial venules (HEV) in peripheral lymphoid tissues.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometric analysis of mouse splenocytes at ≤0.2 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimize antibody titration for best results.

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures designed to minimize spillover and spread.
Excitation: 552 nm; Emission: 592 nm; Laser: 561 nm (Yellow)

Note: Internal testing shows NovaFluor Yellow 590 may non-specifically stain B cells in SJL mice. No such staining observed in BALB/c or C57BL/6 strains.

Target Information

Human CD62L (L-Selectin) is a 74–95 kDa glycoprotein adhesion receptor with:

• C-type lectin binding domain
• EGF-like domain
• Two short consensus repeat (SCR) sequences
• Transmembrane and cytoplasmic regions

CD62L is constitutively expressed on most leukocytes including lymphocytes, monocytes, and polymorphonuclear cells (except a subset of memory T cells).
