Product Specific Information

Description:
The MEL-14 monoclonal antibody reacts with mouse CD62L, a 76 kDa member of the selectin family. CD62L is expressed by neutrophils, monocytes, and subsets of T, B, and NK cells, and binds several glycosylated, fucosylated, and sulfated sialylated glycoproteins including CD34, glycam-1, and MAdCam-1. These interactions mediate lymphocyte rolling on activated endothelium and homing to high endothelial venules (HEV) of peripheral lymphoid tissues.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Use ≤ 0.2 µg per test (100 µL final volume). Optimal cell number: 10⁵–10⁸ cells/test. Antibody titration recommended for best performance.

Compatibility Notice:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for best results.

Blocking Buffer Information:
This NovaFluor conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which improves staining performance compared to the original CellBlox.

• Add 5 µL per sample before combining with cells.
• For single-color controls: use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates are based on Phiton™ technology utilizing novel nucleic acid dye structures engineered for improved fluorescent signatures and reduced spillover/spread.

Target Information

Human CD62L (L-Selectin) is a 74–95 kDa glycoprotein member of the selectin family of adhesion receptors. It includes:

• Amino-terminal C-type lectin binding domain
• EGF-like domain
• Two short consensus repeat (SCR) sequences
• Spacer, transmembrane, and cytoplasmic regions

CD62L is constitutively expressed on all leukocytes, including lymphocytes (except many memory T cells), monocytes, and polymorphonuclear cells.
