Product Specific Information
- Immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
- Staining of formalin-fixed tissues requires heating sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 min.
Target Information
Protein p21 is a tumor suppressor protein that inhibits Cyclin-dependent kinases (CDK) and is transcriptionally activated by p53.
It plays a key role in cellular responses to genotoxic stress and is a major transcriptional target of p53.
In the nucleus, p21 binds and inhibits Cdk1 and Cdk2, blocking the transition from G1 to S phase or G2 to mitosis after DNA damage, allowing DNA repair.
In the cytoplasm, p21 exhibits anti-apoptotic activity by inhibiting caspase 3, ASK1, and JNK.
Thus, p21 has dual roles in carcinogenesis, acting both as a tumor suppressor and as an oncogene.
