Target Information

Sonic Hedgehog (SHH), expressed during embryogenesis, plays a critical role in patterning the early embryo. It is a key inductive signal in ventral neural tube formation, limb axis development, and somite patterning. The human SHH protein is most similar to the Drosophila counterpart. It is synthesized as a precursor that undergoes autocatalytic cleavage:

  • N-terminal portion: Soluble and responsible for signaling activity
  • C-terminal portion: Involved in precursor processing and attaches a cholesterol moiety to the N-terminal product, anchoring it to the cell surface

Defects in SHH or its pathway can cause holoprosencephaly (HPE), characterized by improper forebrain division and facial deformities, and may also contribute to VACTERL syndrome involving multiple congenital anomalies.
