Target Information
GAD-65 and GAD-67 are members of the group II decarboxylase family of proteins and catalyze the rate-limiting step in GABA (gamma-aminobutyric acid) synthesis from L-glutamic acid.
Both are found in the brain:
- GAD-65 localizes to synaptic vesicle membranes in nerve terminals.
- GAD-67 is distributed throughout the cell and maintains basal GABA synthesis.
Under increased neurotransmission demand, GAD-65 transiently activates to assist GABA production. Loss of GAD-65 impairs neurotransmission, while loss of GAD-67 is lethal.
Alternative splicing produces two isoforms: predominant GAD-67 and minor GAD-25.
