Target Information
The gene encodes a member of the tumor necrosis factor (TNF) cytokine family, serving as a ligand for osteoprotegerin and playing a key role in osteoclast differentiation and activation. It functions as a dendritic cell survival factor and regulates T cell-dependent immune responses. T cell activation induces expression of this gene, promoting osteoclastogenesis and bone loss. The protein activates antiapoptotic kinase AKT/PKB via a signaling complex involving SRC kinase and TRAF6, implicating a role in apoptosis regulation. Knockout mice exhibit severe osteopetrosis, absence of osteoclasts, and defects in T/B lymphocyte differentiation and mammary gland development. Two alternatively spliced transcript variants have been identified.
