Target Information
Ago 단백질은 대부분의 체세포에서 발현되며, miRNA와 결합하여 다양한 RNA 침묵화 경로에서 핵심 역할을 합니다. 그중 Ago2 단백질만이 endonucleolytic 또는 “Slicer” 활성을 가지며, miRNA와 완전 상보적인 mRNA를 절단할 수 있습니다. 부분 상보적인 경우에는 절단 대신 번역 억제를 유도합니다.
Ago2는 RNA 간섭, 번역 억제, 이질염색질화 등 작은 RNA 유도 유전자 침묵화 과정에서 비중복적 역할을 하며, RISC 복합체의 핵심 요소로 작용하여 siRNA나 miRNA에 의해 유도된 유전자 침묵화 과정에서 mRNA 분해를 촉매합니다. 또한 Dicer 비의존적 RNA 절단 및 miRNA 성숙 조절에도 관여합니다. Ago2의 과발현은 암세포 성장 및 예후와 관련이 있으며, 마우스 모델에서 Ago2 유전자의 결손은 배아 발달에 필수적임이 밝혀졌습니다.
