Target Information
ADP-ribosylation factors (ARFs)는 RAS 초가족에 속하는 저분자량 GTP 결합 단백질입니다. 예측된 182 아미노산으로 구성된 ARL3(ADP-ribosylation-like factor) 단백질은 rat ARL3와 97%, human ARF1과 43%의 아미노산 서열 유사성을 가집니다. ARFs와 마찬가지로 ARL3는 N-myristoylation 부위인 2번 위치에 glycine을 가지며, RAS 계열의 다른 단백질에서 발견되는 C-말단 근처의 cysteine 잔기가 없습니다. Northern blot 분석에서는 모든 조직에서 1-kb ARL3 전사체가 검출되었으며, 심장과 폐에서 가장 높은 발현을 보였습니다. 5.5-kb 전사체는 대부분의 조직에서 검출되었고, 특히 뇌에서 높은 발현을 나타냈습니다. Immunoblot 분석에서는 ARL3가 인간 종양 세포주에서 검출되었으나 정상 설치류 세포에서는 검출되지 않았습니다. ARL3는 GTP에 결합하지만, cholera toxin 의존적 ADP-ribosylation 반응에서는 활성이 없어 ARF-like 단백질로 분류됩니다.
