Target Information
ADP-ribosylation factors (ARFs)는 RAS superfamily에 속하는 저분자량 GTP 결합 단백질입니다. ARL3 (ADP-ribosylation like factor)는 182개의 아미노산으로 구성된 단백질로, rat ARL3와 97%, human ARF1과 43%의 아미노산 서열 유사성을 가집니다. ARL3는 N-myristoylation 부위에 해당하는 glycine을 2번째 위치에 가지며, RAS family의 다른 단백질에서 발견되는 C-말단 근처의 cysteine 잔기를 결여하고 있습니다. Northern blot 분석에서 ARL3 전사는 대부분의 조직에서 검출되었으며, 심장과 폐에서 가장 높고, 뇌, 간, 신장, 난소, 고환 등에서는 낮은 수준으로 발현되었습니다. 또한, 5.5-kb 전사체는 대부분의 조직에서 검출되었으며 특히 뇌에서 높게 나타났습니다. Immunoblot 분석에서는 인간 종양 세포주에서 ARL3가 검출되었으나 정상 설치류 세포에서는 검출되지 않았습니다. ARL3는 GTP를 결합하지만 cholera toxin 의존적 ADP-ribosylation 반응에는 관여하지 않아 ARF-like 단백질로 분류됩니다.
