Target Information
CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase (PTP) expressed in all nucleated hematopoietic cells, comprising approximately 10% of all surface proteins in lymphocytes. It is absent in non-hematopoietic cells and tissues. CD45 plays a crucial role in lymphocyte development and antigen signaling by regulating Src-family kinases.
Multiple isoforms of CD45 exist due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic regions. CD45RA is one such isoform with restricted expression among lymphoid cell subtypes.
CD45 regulates T- and B-cell receptor signaling, suppresses JAK kinases in cytokine signaling, and modulates integrin-mediated pathways affecting cell survival and apoptosis.
