Description:
The eBioMWReg30 monoclonal antibody reacts with mouse CD41 (fibrinogen receptor, gpIIb, integrin alpha IIb). CD41 is expressed on platelets, megakaryocytes, and hematopoietic progenitors in embryo, fetus, and adult. It associates with CD61 (gpIIIa) to form a receptor involved in platelet function and adhesion molecule binding (fibrinogen, fibronectin, vitronectin).

CD41 expression is also relevant for hematopoietic stem cell (HSC) identification. CD150⁺CD48⁻CD41⁻ cells efficiently identify long-term multi-lineage reconstituting HSCs.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of mouse platelets. Recommended ≤0.125 µg/test (in 100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimize by titration for best performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining with NovaFluor conjugates.
• Add 5 µL of CellBlox Plus per sample (regardless of number of NovaFluors).
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Based on Phiton™ technology using nucleic acid dye structures for engineered fluorescent signatures with minimized spillover and spread.

Target Information

CD41 (platelet glycoprotein IIb, ITGA2B) consists of α (120 kDa) and β (23 kDa) subunits that interact with CD61 in the presence of calcium to form a functional adhesive receptor. It mediates platelet aggregation and binds von Willebrand factor, fibrinogen, fibronectin, and vitronectin. Expressed mainly in megakaryocyte-platelet lineage and early hematopoietic differentiation stages.
Associated diseases include Glanzmann Thrombasthenia and Platelet type-16 bleeding disorder.
