Product Specific Information

The RPA-T4 monoclonal antibody reacts with human CD4, a 59 kDa cell surface receptor expressed by most thymocytes, a subpopulation of mature T-helper cells, and at low levels on monocytes. CD4 acts as a receptor for human immunodeficiency virus (HIV). RPA-T4 blocks HIV binding and mixed lymphocyte reactions and recognizes a different epitope than OKT4, with no cross-blocking between the two.

  • Applications Reported: Flow cytometric analysis
  • Applications Tested: Pre-diluted and tested for flow cytometry using normal human peripheral blood cells.
    • Use at 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorochrome Information:
Brilliant Ultra Violet™ 615 (BUV615) emits at 615 nm and is suitable for cytometers equipped with a 355 nm UV laser. Ensure instrument compatibility.

Buffer Recommendations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Light Sensitivity:
Protect from light to prevent photo-induced oxidation.

Target Information

CD4 is involved in MHC class II recognition and functions as a co-receptor for HIV. It is primarily expressed on T-helper lymphocytes and also found on monocytes, macrophages, and dendritic cells. Tissue expression includes thymus, lymph nodes, tonsils, spleen, and certain brain and gut regions. CD4 facilitates T-cell activation via association with the TCR complex and protein tyrosine kinase Lck, and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
