Target Information

Phosphatidylinositol (3,4,5)-trisphosphate (PtdIns(3,4,5)P3), commonly abbreviated as PIP3, is the product of class I phosphoinositide 3-kinases (PI3Ks) acting on phosphatidylinositol (4,5)-bisphosphate. PtdIns(3,4,5)P3 is dephosphorylated by PTEN on the 3-position and by SHIPs (SH2-containing inositol phosphatases) on the 5′ position of the inositol ring.

The PH domain in various proteins binds to PtdIns(3,4,5)P3, including Akt/PKB, PDK1, Btk1, and ARNO. Generation of PtdIns(3,4,5)P3 at the plasma membrane upon activation of class I PI3Ks causes these proteins to translocate to the membrane, affecting their activity. The PH domain also mediates binding between PtdIns(3,4,5)P3 and G-protein coupled receptor kinases (GRKs), enhancing GRK association with the plasma membrane.
